We need more of this kind of studies. We always get single papers where they show how incredibly efficient their methods is on the couple of loci they selected. To date I do not know any genome engineering technique that works efficiently in all loci #CRIS

We used a plasmid that had both loxP sites surrounding knockout region (~2kb) with 2.5kb HAs and a single gRNA. Worked well for us.
